摘要
目的探讨使人骨髓间充质干细胞(MSCs)向成骨细胞诱导分化的高效诱导体系。方法按照不同浓度的地塞米松、β-甘油磷酸钠和维生素C分将实验为4组,并设立空白对照组,诱导人骨髓间充质干细胞向成骨细胞定向分化,应用碱性磷酸酶钙钴法染色检测分化的成骨细胞的成骨特性。结果在地塞米松10-8mol/L、β-甘油磷酸钠10-2mol/L、维生素C 3×10-4mol/L时,积分光密度值最高。结论 MSCs具备较强的增值能力和良好的成骨特性,可为骨缺损修复技术优化提供基础。
Objective To explore efficient induction system of inducing marrow mesenchymal stem cells to osteoblasts.Methods Trial was divided into 4 groups according to different concentrations of vitamin C,dexamethasone and β-glycerophosphate,and the control group was set.Human marrow mesenchymal stem cells were induced into osteoblasts,then the characteristic of osteoblasts was detected by gomori method for alkaline phosphatase(ALP).Results Under the concentration of dexamethasone of 10-8mol/L,β-glycerophosphate of 10-2mol/L and vitamin C of 3×10-4mol/L,integrated optical density was the highest.Conclusion Mesenchymal stem cells poses potential augment capability and excellent osteogenic characteristics,which provides foundation for technique optimization of bone defeat fixation.
