德国骑乘马线粒体DNA的限制性酶分析

Restriction Endonuclease Analysis of Mitochondrial DNA of German Saddle Horse

通过５种限制性内切酶获得了１３匹德国骑乘马线粒体ＤＮＡ（ｍｔＤＮＡ）的切割图谱。结果发现，限制性消化产生了１（ＥｃｏＲⅠ），３（ＢｇｌⅡ），４（ＢａｍＨⅠ）或５个（ＨｉｎｄⅢ和ＢａｍＨⅠ）限制性片断，并确定了所有片断的分子大小。ｍｔＤＮＡ总的大小平均为（１６．４８±０．１０）ｋｂ．除ＢａｍＨⅠ外，ｍｔＤＮＡ的限制性切点对于所有的个体是完全一致的。用ＢａｍＨⅠ检测到ｍｔＤＮＡ的多态性，通过带谱分析推断，德国骑乘马的Ｂ型ｍｔＤＮＡ是由Ａ型ｍｔＤＮＡ通过突变产生１个新的切割位点进化而来。

German saddle horses cleavage patterns of mtDNA from 5 restriction endonucleases ( Bam HⅠ, Hin dⅢ, Eco RⅠ, Eco R V and Bgl Ⅱ) were obtained.The restriction digests yielded one ( Eco R Ⅴ ),3 ( Bgl Ⅱ),4 ( Bam HⅠ) or 5 ( Hin dⅢ and Bam HⅠ) fragments and all fragment sizes were given.The mean total size of mtDNA was calculated to be (16.48±0.10) kilobases.The restriction cleavage sites of mtDNA for all individuals except Bam HⅠ were completely identical.A polymorphism of mtDNA was detected with Bam HⅠ.The kind of polymorphism let us assume that B type mtDNA of this horse had evolved from the A type mtDNA by acquiring an additional cleavage site.
