BN大鼠玻璃体内注射曲安奈德抑制脉络膜新生血管生长 被引量:3

Inhibition of intravitreal injection of triamcinolone acetonide on growth of choroidal neovascularization in BN rats

摘要 目的观察曲安奈德(triamocinlone acetonide,TA)对氪激光诱导的BN大鼠脉络膜新生血管(choroidal neovascularization,CNV)生长的抑制作用。方法 将36只健康BN大鼠随机分为TA组和对照组,每只大鼠随机抽取一眼进行实验。建立BN大鼠CNV动物模型,TA组视网膜光凝后立即玻璃体内注射40g.L-1TA8μL,对照组则注射同等容积的等渗BSS液。在术后2周和4周行FFA检查,FFA检查后摘除BN大鼠眼球制做病理组织切片,光镜下观察CNV情况,并通过免疫组织化学方法检查ICAM-1蛋白在BN大鼠CNV中的表达,采用原位杂交方法检查ICAM-1mRNA在CNV中的转录情况,同时对实验数据进行半定量分析。结果 TA组的ICAM-1蛋白和ICAM-1mRAN阳性着色的IOD值、CNV面积及FFA检测的IOD值明显低于同时期的对照组(均为P<0.05)。结论 TA能够降低ICAM-1mRNA转录和ICAM-1蛋白的表达,有效地抑制了BN大鼠CNV的生长,表明TA是抑制CNV生长的一种很有发展前景的药物。 Objective To observe the inhibition of krypton laser induced choroidal neovascularization(CNV) of BN rat by intravitreal injection of triamcinolone acetonide(TA).Methods Thirty-six healthy Brown Norway(BN) rats were divided randomly into TA group and control group,one eye was chosen randomly as experimental eye.After establishing CNV models of BN rats,40 g·L-1(8 μL) TA was injected into vitreous immediately after photocoagulation in TA group,the same volume isotonic balanced salt solution(BSS) was injected in vitreous in the control group.FFA examination was made at postoperative 2 weeks and 4 weeks.Eyeballs of BN rats were enucleated for histological slices.Situation of CNV was observed under light microscope.The protein expression of intracellular adhesion molecule-1(ICAM-1) of BN rats was observed by immunohistochemical method,and the transcription of ICAM-1 mRNA in CNV was detected by in situ hybridization,meanwhile experimental data were taken semi-quantitative analysis.Results In TA group,the positive staining integrated optical densities of ICAM-1 protein and mRAN,CNV area and FFA integrated optical densities were obviously lower than that in contemporaneous control group(All P0.05).Conclusion TA can lower the expression of ICAM-1 mRNA transcription and the protein expression of ICAM-1,and effectively inhibit the development of CNV in BN rats,which indicate that TA is a prospective drug to block the CNV development.
