兔颅脑爆炸伤后水通道蛋白-4表达与高压氧治疗作用 被引量:11

Expression of AQP-4 and efficacy of hyperbaric oxygen therapy after blast-related brain injury in rabbits

摘要 目的研究兔颅脑爆炸伤后水通道蛋白4(AQP-4)的表达及高压氧早期治疗的影响。方法将新西兰大白兔150只,随机分为对照组、治疗组和非治疗组;其中对照组10只,另两组各70只。非治疗组和治疗组均采用纸雷管爆炸制作兔颅脑爆炸伤模型,并对治疗组采用早期高压氧治疗。三组均采用干湿法测定脑组织含水量,Western Blot法测定AQP-4表达情况。结果非治疗组与治疗组脑组织含水量和AQP-4在伤后1 h开始升高,72 h达高峰,7 d后逐渐下降。在伤后6 h后各时间点,治疗组AQP-4的表达均较非治疗组明显下调(P<0.05)。非治疗组脑组织含水量与AQP-4表达变化呈正相关(r=0.876 7,P<0.01)。结论兔颅脑爆炸伤后AQP-4表达可能与脑组织缺氧有关,而高压氧早期治疗可缓解脑组织缺氧引起的继发性脑水肿。 Objective To investigate the expression of aquaporin-4(AQP-4) and the effect of early hyperbaric oxygen(HBO) on model of blast-related(explosive) brain injury in rabbits.Methods One hundred and fifty New Zealand rabbits were randomly divided into control group(10 rabbits),non-treatment group(70 rabbits) and treatment group(70 rabbits).Explosive brain injury models were made by paper detonator in treatment and non-treatment groups,and HBO therapy was performed only in treatment group.The water content of the brain tissue was detected by dry-wet method in the three groups,and the expression of AQP-4 in the brain tissue was detected by Western blot.Results The water content of the brain tissue and the expression of AQP-4 both began to increase 1 h after the explosive injury,reached the peak at 72 h,and decreased slowly after 7 d in treatment and non-treatment groups.In the time points of 6 h after the explosive injury,the expression of AQP-4 in the treatment group was significantly down-regulated compared with the non-treatment group(P0.05).There was positive correlation between the water content of the brain tissue and the expression of AQP-4 in non-treatment group(r=0.876 7,P0.01).Conclusions The expression of AQP-4 after explosive injury may be associated with cerebral hypoxia,whereas,early HBO therapy could relieve brain edema secondary to brain tissue hypoxia.
