摘要
目的观察血府逐瘀汤复方对高血压血瘀证患者血清致血管内皮功能障碍的影响及机制。方法人脐静脉内皮细胞株ECV-304分组后以MTT法选取该复方药物血清的最佳作用浓度,检测ET和NO、内皮细胞蛋白C受体(EPCR)、血管内假性血友病因子(vWF)和血栓调节蛋白(sTM)的含量。在激光扫描共聚焦显微镜下观察其对细胞内Ca2+浓度(〔Ca2+〕i)和细胞骨架微丝结构的影响。结果与模型组相比,血府逐瘀汤组(〔Ca2+〕i)浓度升高,NO、EPCR和vWF降低,骨架微丝少但排列有序。结论该复方可以有效对抗高血压血瘀证患者血清干预ECV-304造成的内皮功能障碍,从药物的角度反证了细胞损伤模型的建立。
Objective To observe the protective effects of Xuefu Zhuyu decoction on the cellar injury model by the serum of patients with blood stasis syndrome (BSS) associated with hypertension on endothelia dysfunction. Methods Studies were conducted in ECV-304, and they were equally conventionally divided into normal control ( cultured with DMEM), model ( with the serum of patients with BSS associated with hypertension) and medicine groups (with the serum of patients with BSS associated with hypertension and the mice serum of Xuefu Zhuyu decoction). All groups were measured. Results There was significant difference(P 〈 0. 001 )of the secretion of NO, EPCR, vWF, intracellular calcium concentration and the eytoskeleton, between model and medicine groups. Conclusions The Xuefu Zhuyu decoction has the protective effects on endothelial dysfunction of the cellar injury model by the serum of patients with BSS associated with hypertension and from the point of medicine, they could recover endothelial dysfunction induced by cellar injury model.
