富血小板血浆对脂肪间充质干细胞生物学行为的影响 被引量:4

摘要 目的探讨富血小板血浆(PRP)对人脂肪间充质干细胞(hADSCs)在体外培养条件下增殖、分化及细胞成骨能力的影响,研究PRP的生物学功能。方法健康志愿者静脉血经2次离心法制备PRP,氯化钙加入凝血酶激活后提取萃取液。hADSCs成骨诱导实验分4组,即空白对照组、只加成骨诱导液组、只加50%PRP 20μl组、既有成骨诱导液又加50%PRP 20μl组,倒置相差显微镜观察细胞增殖、分化。碱性磷酸酶(ALP)染色实验分4组,即空白对照组、不同浓度PRP(即原液、50%、25%)组,每孔各加20μl PRP,第6天ALP活性染色。细胞增殖能力检测,成骨诱导培养后2、4、6、8、16 d采用CCK-8法检测不同浓度PRP对细胞增殖活性的影响。结果成骨诱导实验中倒置相差显微镜见加PRP组细胞数量多于单纯诱导组和空白组,细胞形态多数为梭形;ALP活性检测见高浓度PRP组ALP阳性细胞深染,并有浓度依赖性。CCK-8测定细胞增殖活性,显示12.5%PRP组第4、6天平均OD值分别为0.035、-0.063,高于对照组(-0.833、-1.041)(P<0.05)。结论适宜浓度的PRP可促进hADSCs的增殖,但对其成骨分化有抑制作用。过高浓度的PRP虽然同样有促进增殖的作用,但并非浓度越高越好。
