摘要
目的:研究影响细支气管肺泡癌(BAC)组织中凋亡相关基因Bcl-2、bax和Caspase-3表达的相关因素。方法:收集经病理学确诊且临床资料完整的BAC患者38例,RT-PCR法测定其肿瘤组织中Bcl-2、bax和Caspase-3 mRNA水平,并分析其与BAC患者性别、吸烟史、肿瘤临床分型及TNM分期之间的关系。结果:女性BAC组织的Bcl-2mRNA水平显著高于男性,P<0.001,而Bax和Caspase-3 mRNA水平男、女性间差异无统计学意义,P>0.05;吸烟者BAC组织的Bcl-2 mRNA水平显著低于不吸烟者,P<0.001,而Bax和Caspase-3 mRNA水平显著高于不吸烟者,P均<0.001;孤立结节型、多发结节型、肺炎型BAC组织的Bcl-2 mRNA水平依次显著增高,P<0.001,而Bax和Caspase-3 mRNA水平依次显著下降,P均<0.001;随着TNM分期由Ⅰ期至Ⅳ期的逐级升高,BAC组织的Bcl-2 mRNA水平依次显著增高,P<0.001,而Bax和Caspase-3 mR-NA水平依次显著下降,P均<0.001。结论:BAC组织中凋亡相关基因Bcl-2、Bax、Caspase-3 mRNA水平与肿瘤患者的性别、吸烟史、临床分型、TNM分期均有不同程度和方向的相关性。
OBJECTIVE: To study the factors related with the expression of apoptosis-related genes(gcl-2, bax, Caspase-3) in bronchioloalveolar carcinoma(BAC). METHODS: Totally 38 pathologically diagnosbd BAC cases were collected. The levels of Bcl-2, bax and Caspase-3 mRNA in the carcinomas were deter- mined, and the relationships of the levels with the gender, smok- ing, clinical type and TNM stage were analyzed statistically. RE- SULTS: The Bcl-2 mRNA of female group was significantly high- er than that of male group (P〈0. 001), but there were no signif- icant differences between the two groups in Bax and Caspase-3 mRNA levels (P〈0.05). The Bcl-2 mRNA of smoking group was significantly lower than that of unsmoking group (P 0. 001), but the Bax and Caspase-3 mRNA levels were significantly higher than those of unsmoking group (P〈0. 001). The Bcl-2 mRNA levels of solitary nodular type, multiple nodular type and pneumonia type were significantly increased by turns (P〈0. 001), but the levels of Bax and Caspase-3 mRNA were significantly decreased by turns (P%0. 001). With the stage of the carcinoma advancing from Ⅰ to Ⅳ, the Bcl-2 mRNA levels significantly increased by turns (P〈0. 001), hut the the levels of Bax and Caspase-3 mRNA were significantly decreased by turns (P〈0. 001). CONCLUSION: There exist correlations of different grades between the expression of apoptosis-related genes (Bcl-2, Bax, Caspase-3) mRNA and the gender, smoking, clinical type and TNM stage in bronchioloalveolar carcinoma.
