摘要
从ITS序列和ISSR分子标记水平对45株棉花黄萎菌进行遗传变异研究,共扩增到178条多态性条带,呈现丰富的多态性。依据ISSR分子标记的聚类分析结果、菌株的致病性以及ITS测序结果,可将45个菌株分为弱致病类型大丽轮枝菌、强致病类型大丽轮枝菌和变黑轮枝菌3类,ISSR分子标记多态性与棉花黄萎病菌的致病类型呈现一定的相关性,但与保存年限间无明显的相关性,所有菌株均表现出了典型的分子标记模式,仅有菌株394的ITS序列出现了单位点突变。
The genetic variation analysis of 45 isolates of Verticillium wilt pathogen was conducted through ITS(internal transcribed spacer) sequences and ISSR(internal simple sequence repeat) markers.One hundred and seventy-eight polymorphism bands were obtained from ISSR markers.Based on the clustering result of ISSR marker data,pathogenicity and ITS sequence of the strains,forty five isolates were divided into three groups: V.dahliea with weak virulence,V.dahliea with strong virulence,and V.nigrescens.The polymorphism of ISSR has some kind of correlation with pathogenicity,but no obvious relationship with the preserved years of the strains.All the isolates present the typical molecular marker pattern,except that the strain 394 appeares single site mutation.
