阉割对金华猪甲状腺组织GNAS基因表达的影响

摘要 为研究阉割对甲状腺刺激性G蛋白α亚基(GNAS)基因表达的影响,采用SYBR Green荧光定量PCR分析方法,分析60、90和120日龄阉割组与非阉割组金华猪甲状腺组织中GNAS基因的表达水平。结果表明:在3个时期,无论是阉割组还是非阉割组,金华猪GNAS基因的表达水平均是90日龄低于60日龄,而120日龄又高于90日龄;非阉割组60日龄GNAS基因的表达量最高,而阉割组120日龄GNAS基因的表达量最高。阉割组GNAS基因的表达水平在不同的生长阶段均低于非阉割组。结果提示建立的SYBR-Green荧光定量PCR方法可有效地用于GNAS基因的表达分析;阉割会造成猪GNAS基因的表达量下降。 In order to investigate the association of castration with the expression of guanine nucleotide-binding protein alpha-stimulating activity polypeptide(GNAS) gene,the SYBR-Green real-time PCR method was established and used to explore expression of GNAS gene from the thyroid tissue of castrated and uncastrated Jinhua pigs at the age of 60,90 and 120 days,respectively.The results showed that GNAS gene expression in day 90 was lower than day 60,at the same time the expression in day 120 was higher than day 90 in castrated and uncastrated group in all stages;the expression of GNAS gene in day 60 was the highest in uncastrated group,while in castrated group it is the highest in day 120.The GNAS expressions in castrated Jinhua pigs were lower than those of uncastrated pigs at all three stages examined.These results suggested that the real-time PCR protocol established is available for the expression analysis of porcine GNAS gene,and that the castration may decrease the expression of the GNAS mRNA in pigs.
