摘要
目的建立大鲵基因组DNA快速简便高效的提取方法。方法借鉴煮沸裂解法制备质粒DNA方法,改进后用于提取大鲵基因组DNA。结果本方法提取DNA纯度高(A260/A28介于1.7~2.1之间),耗时短(2h),毒性小,分子大小约为23kb,适用于PCR扩增及后续分析。结论本方法提取基因组DNA纯度高、耗时短、提取所用试剂毒性小、成本低,值得在大鲵基因组DNA提取中使用,也可为其他动物的相关研究提供参考。
Objective To found a quick,simple,efficient separation method of Giant Salamander genome DNA.Methods Based on the boiling lysis method of plasmid DNA separation,improved the protocol for Giant Salamander genome DNA extraction.Results The A260/ A280 of DNA extracted from Giant Salamander was pure highly(between 1.7 and 2.1).It took very short time(2h) and have low toxicity of reagent.The fragment of extracted DNA was about 23kb and it was suitable for PCR and subsequent analysis.Conclusions This method was at high pure,short time,harmless,low cost,and fit for extracting of Giant Salamander genome DNA,and also be used to similar studying in other animals.
