端粒酶反义寡聚脱氧核苷酸对HL-60细胞生长的作用

The effect of telomerase antisense oligodeoxynucleotides on the growth of HL-60 cell line

目的：观察端粒酶反义寡聚脱氧核着酸对急性髓细胞白血病细胞株HL-60的作用、方法：采用台盼蓝排斥法进行药物敏感试验，流式细胞仪测定凋亡细胞含量结果：端粒酶反义寡聚脱氧核着酸对HL-60细胞有生长抑制作用，作用具有序列特异性及剂量依赖性．流式细胞仪检测到凋亡峰，含量为28．8％．无关寡聚核着酸片段无此作用．结论：端粒酶对维持白血病细胞的生长可能起十分重要的作用，它有可能成为白血病治疗的新靶点．

AIM: To observe the effect of telomerase antisense oligodeoxynucleotides on HL 60 cell line. METHODS:The drug sensitivity test was performed with trypan blue and the apoptotic cells were measured with flow cytometer (FCM ). RESULTS: Telomerase antlsense oligodeoxynu cleotides acted as a specific growth Inhlbltor to HI- 60 cell line and the effect was in sequence specific and dose dependent manner. The apoptotic peak was detected with F(tM and the quantity was 28. 8%. Non--oligodeoxynucleotlde had no inhibitory effect on HL 60 cell line. CONCLUSION: Telomerase may play a crucial role in the growth of leukemic cells.It may be a new target of treatment to leukemia.
