局部加温与紫杉醇对小鼠肺腺癌LA_(795)抑瘤增效的研究 被引量:1

摘要 目的观察局部加温与紫杉醇对小鼠肺腺癌LA795的联合治疗作用。方法将瘤细胞悬液接种于小鼠右后肢足背皮下,待肿瘤平均瘤积为110mm3时,平均分组,根据实验要求分别进行紫杉醇治疗、热疗、联合治疗处理。结果当小鼠平均瘤积V≥1000mm3和2000mm3时,联合治疗组的肿瘤生长延迟时间(TGD)为3天,肿瘤抑制率为58%(t=6.8,P<0.001)和40%(t=3.1,P<0.01)。q值分别为1.50和1.38。光镜观察,肿瘤组织以广泛的凝固性坏死,肿瘤细胞出现重度空泡变性坏死,间质纤维组织增生,微血管高度扩张,小静脉瘀血,皮下与肿瘤之间肉芽组织膜形成并见少量淋巴细胞和浆细胞。电镜观察,肿瘤细胞核变性,溶酶体多,细胞器变性,粗面内质网脱粒,膜系统不同程度的破坏等。结论局部加温与紫杉醇联合治疗对肿瘤组织显示协同杀伤作用,具有临床应用价值。 Objective To investigate whether the paclitaxel and local thermotherapy combining therapy can delay the growth of lung adenocarcinoma of mice.Methods A suspension of LA 795 lung adenocarcinoma cells was implanted into the right hind leg of each mouse subcutaneously.When the volume of tumor was reaching 110 mm 3 on average,these mice were divided into the paclitaxel group,paclitaxel and thermotherapy combining group,thermotherapy group,control group.Each group was 10 mice.Results On the tumor volume 1000 mm 3 and 2000 mm 3,the time of tumor growth delay(TGD) in combining therapy group was averagely 3 days and their inhibitive rate was 58% and 40% respectively.By light microscope examination,the wide-spread coagulation necrosis,vacuole degeneration of cancer cell,interstitial fibrous hyperplasia,microvascular dilatation and vanulae bloodstasis in tumor tissues were mostly presented in combining therapy group compared with other groups.By electron microscope,the nucleus degeneration of cancer cell,hyper-lysosome,cytoplation of rough endoplasmic reticulum and membrane damage were obviously demonstrated in combining therapy,too.Conclusion It is showed that the paclitaxel plus thermotherapy is an effctive method for lung cancer,it has a 3cooperation for killing the LA 795 lung adenocarcinoma of mice.
