摘要
目的观察肿瘤转移相关基因(MTAl)在原发性肝细胞癌(HCC)中的表达,探讨MTA1与HCC浸润和转移的关系。方法分别采用免疫组织化学sP法和逆转录-聚合酶链反应(RT—PCR)方法检测42例HCC、10例肝硬化和7例正常肝组织内MTA-1的表达,并结合临床病理指标进行分析。结果MTAl在HCC、肝硬化和正常肝脏组织中的阳性例数分别为26、2和0例,差异有统计学意义(P〈0.05);HCC组织内MTAlmRNA的表达强度明显高于肝硬化组织(t=2.792,P〈0.05);MTAl在有包膜侵犯组、病理分级Ⅲ~Ⅳ组及有转移HCC中的阳性表达率及强度明显高于无包膜侵犯组(P〈0.05)、病理分级I~Ⅱ级组(P〈0.05)以及无转移组(P〈0.01)。结论MTAl在HCC组织中的表达上调与HCC的浸润和转移有关。
Objective To study the expression of metastasis associated-1 ( MTA1 ) in human hep- atocellular carcinoma (HCC) and its relationship with invasion and metastasis of HCC. Methods By u- sing the method of immohistochemical staining (SP) and reverse transcription-polymerase chain reaction (RT-PCR) , specimens from 42 cases of HCC, 10 cases of hepatic cirrhosis (HC) and 7 normal liver tis- sues (NL) were studied. The relationship between the MTA1 expression and HCC pathological features was evaluated. Results MTA1 positive cases in the HCC, HC and NL tissues was 26, 2 and 0 respectively, with the significant difference between HCC and HC as well as NL ( P 〈 O. 05 ). The expression of MTA1 mRNA in HCC tissues was significantly higher than that in HC ( t = 2. 792, P 〈 O. 05 ). Overexpression of metastasis associated-1 in the HCC was related to the invasion of tumor envelop ( P 〈 0. 05 ) , Edmondson grades (P 〈 0. 05 ) and tumor metastasis (P 〈 0. 01 ). Conclusion MTA1 may play an important role in invasion and metastasis of HCC.
