开心解郁汤对抑郁模型大鼠脑皮质内单胺类神经递质和行为学的影响 被引量:7

Effects of Kaixin Jieyu Decoction on the Monoamines Neurotransmitter in the Brain Cortex and Behaviors of Depressed Rat Model

摘要 目的:研究开心解郁汤对抑郁模型大鼠脑皮质中单胺类神经递质含量和行为学能力的影响,探讨开心解郁汤治疗抑郁症的可能机制。方法:采用慢性不可预见性温和应激方法(CUMS)造成抑郁大鼠模型,随机分组分别灌胃给予生理盐水、开心解郁汤和盐酸氟西汀。22d后分取脑皮质,HPLC-ECD法测定脑皮质中5-HT和NE等单胺类神经递质的含量,并观察大鼠行为学变化。结果:模型组大鼠脑皮质中5-HT、NE水平下降(P<0.01,P<0.05),体重增加数和糖水消耗量均下降,旷场实验测定水平活动和垂直活动次数均下降(P<0.01,P<0.05);经开心解郁汤和盐酸氟西汀治疗后大鼠脑皮质中5-HT、NE水平明显上升(P<0.01,P<0.05),糖水消耗量、水平活动和垂直活动次数均较模型组明显增加(P<0.01,P<0.05)。结论:CUMS可引起脑皮质内单胺类神经递质(5-HT、NE)含量和大鼠行为学变化,造成大鼠抑郁模型。开心解郁汤可以显著改善抑郁大鼠脑内单胺类神经递质水平和行为能力。 Objective:Investigate the effect of Kai Xin Jie Yu Decoction on the monoamines neurotransmitter in brain cortex and behaviors of rats depression model,and study the anti-depression mechanism of Kai Xin Jie Yu Decoction.Methods:The rats depression model was established by chronic unpredictable mild stress(CUMS).SD rats were divided randomly into normal,depression,depression + herbal medicine,and depression + Fluoxetine group,and were administrated with normal saline,Kai Xin Jie Yu Decoction,and Fluoxetine Hydrochloride separately.The model was evaluated regularly with monoamines neurotransmitter in the brain cortex,the weight,and the consumption of hedonia,and the horizontal and vertical activity(Open-field test).Results:Compared with normal group,5-HT and NE in brain cortex,the weight increase,the consumption of hedonia and number of the horizontal and vertical activity were decreased in the depression model group.While increased obviously in the Kai Xin Jie Yu Decoction group and Fluoxetine Hydrochloride treatment group.Conclusion:The rats depression model could be established by CUMS.5-HT and NE in the brain cortex and behaviors of the rats depression model were changed.The monoamines neurotransmitter in brain cortex and behaviors of rats depression model were improved obviously by Kai Xin Jie Yu Decoction.
