葛根素对糖尿病大鼠心肌细胞凋亡及相关蛋白表达的影响 被引量:21

摘要 目的:探讨葛根素(Puerarin,Pue)对糖尿病大鼠心肌细胞凋亡及相关蛋白表达的影响。方法:40只SD大鼠随机分成空白组、糖尿病模型组、Pue低剂量组和Pue高剂量组各10只。对4组大鼠行空腹血糖、心肌细胞凋亡指数(AI)、免疫组化和Western-Bloting检测。结果:Pue各治疗组均较模型组有显著意义改变:空腹血糖降低;AI下降;Bcl-2表达升高,Caspase 3和Cyt-C表达均下降;且高剂量组的改善情况更为理想。结论:Pue可以改善糖尿病大鼠空腹血糖和AI,其机制可能与调节Bcl-2、Caspase 3和Cyt-C水平有关。 Objective:To investigate the effects of Puerarin on myocardial cell apoptosis and related protein expression in diabetic rats.Methods:The forty SD rats were randomly divided into four groups:the control group,the diabetic model group,the Puerarin low-dose group and high-dose group.Rats were respectively measured fasting blood glucose,apoptosis index(AI),immunohistochemistry and Western Blot.Results:Puerarin therapy groups than model group are statistically significant improvement:fasting blood glucose and AI were decreased,Bcl-2 expression in turn is increased,Caspase 3 and Cyt-C expression are decline in turn,and the high doses group improvement situation is most ideal.Conclusion:The expression of Bcl-2、Caspase 3 and Cyt-C are related to diabetic rats,Puerarin has protective effect on blood glucose and AI through affecting the changes of above mentioned indexes.
