摘要
目的研究特发性血小板减少性紫癜(ITP)患者血浆中可溶性CD40配体(sCD40L)及T、B淋巴细胞和血小板上CD40L的变化,探讨CD40分子在ITP中的作用。方法应用酶联免疫吸附法检测患者血浆中sCD40L的变化;流式细胞术检测不同细胞上CD40L的表达水平。结果 ITP患者血浆中的sCD40L浓度明显高于正常对照组(P<0.05),且血小板计数对血浆中sCD40L浓度有一定程度的影响(r=0.58,P<0.05);ITP患者T淋巴细胞上CD40L的表达率较正常对照组明显升高(P<0.05)。结论 ITP患者血浆及T细胞上CD40L升高,淋巴细胞表面的CD40-CD40L相互作用可能与ITP的发病有关。
Objective To investigate the alternate in plasma soluble CD40L ligand(sCD40L),and CD40L on B,T lymphocytes and platelets in idiopathic thrombocytopenia purpura(ITP).Methods The plasma level of sCD40L was measured by ELISA method,and CD40L of other cells were measured by using flowcytometry.Results The plasma level of sCD40L in ITP was higher than that in the normal controls(P0.05).There was significant positive correlation between sCD40L values and platelets counts in ITP patients(r=0.58,P0.05).The expression of CD40L on T lymphocytes and platelets increases in ITP(P0.05).Conclusion The level of CD40L in ITP patients is higher than that in the normal controls in serum and on T lymphocytes.Lymphocytes surface of CD40-CD40L interaction may be related to the pathogenesis of ITP.
