基于二次正交旋转回归优化大豆根内拮抗菌AF-67发酵工艺 被引量:3

Optimization of Fermentation for Antifungal Entophytic Bacteria AF-67 by Quadratic Orthogonal Regression Design

摘要 应用二次正交旋转回归设计,研究培养基关键影响的四因子对大豆根腐生防菌AF-67发酵菌液浓度的影响.优化发酵培养基各成分的最佳配比为:牛肉浸膏1%,蛋白胨3.5%,可溶性淀粉1.8%,葡萄糖1%,发酵24h后稀释10倍工作液的实际光密度值A_(620)为0.743,较LB增殖培养液提高了42%.肯定了模型的可靠性和应用的有效性. The test was designed by using the Quadratic Orthogonal Rotation Combination Design,and the effect of various factors on response value for fermentation broth of AF-67 was analyzed.The results showed that ingredients of fermentation medium were optimized as follows: 1%beef extract,3.5%peptone,1.8%soluble starch and 1%glucose.Perform fermentation test by using the optimized medium,and the result showed that the mean A620 value of fermentation broth was 0.743,which was increased by 42%as compared with the proliferation of LB culture medium.The statistic test showed that the regression model is reliable and valid.
