摘要
目的:研究乳杆菌Lb.casel Zhang对小鼠脾细胞IL-2和其受体IL-2R仪的诱生作用,从基因转录水平探讨其免疫调节的特点。方法:利用分离自酸马奶,并经过耐酸性实验、人工胃肠消化液中存活能力筛选的Lb.caseiZhang,制备活菌制剂分高、中、低三个剂量组灌服小鼠,分别于第3、7、11、15d,摘取脾脏,计算脾指数,RT—PCR法测定睥脏中IL-2和IL-2R仪mRNA的转录水平。结果:脾指数在连续灌服7d后开始明显增加,至11d时仍保持较高水平,并具有一定的剂量依赖效应;IL-2和IL-2R仪mRNA的转录水平与脾指数相适应在连续灌服7d后开始明显增加,至11d时达到高峰,且具有一定的剂量依赖效应。结论:乳杆菌Lb.caseiZhang能够诱导脾脏淋巴细胞自分泌生长因子IL-2及其活化受体IL-2R仅的转录及分泌,进而激活Till细胞,从而发挥免疫调节作用。
Objective: To study the induction IL -2 and IL2Rα receptors by Lb. casei Zhang in splenocyte of mouse; and discuss the characteristics of immunoregulation of Lb. casei Zhang from the perspective of gene transcription level . Methods:Method RT- PCR assay the transcription level of IL - 2, IL - 2Rα and mRNA in respectively collected spleens of mice on the third day, seventh day, eleventh day and the fifteenth day after been orally administrated with three variety of dosages of living Lb. casei Zhang, which were isolated from koumiss and could resist acid artificial gastric and intestinal juice. Results:Spleen index starts to significantly increase after 7days' successive perfusion and remains at a high level on the eleventh day. There is a certain dose - dependent effect. Transcription level of IL - 2 and IL - 2Rα mRNA together with spleen index start to go up after 7days' successive perfusion and remain at a higher level than the control group until the eleventh day. There is a certain dose -dependent effect. Conclusion:Lb. casei Zhang can induce the transcription and secretion of autocrine growth factor IL -2 and activated receptorIL -2Rα in spleen Lymphocytes. And further to activate TH1 cell, to play part in immunoregulation and to promote gut immune cellularity.
