摘要
目的:探讨立体定向移植方法在干细胞移植中的应用。方法:腹腔注射青霉素建立癫痫模型。24只健康Sprague—Dawley SD大鼠分为生理盐水移植组及干细胞移植组。用立体定向仪进行移植,分别于移植后7d脑灌注取材,通过荧光观察比较干细胞移植后干细胞在移植部位的准确性,立体定向移植方法的安全性。结果:发现移植后7d脑片,在针道内均有大量的GFP标记的细胞存在,且无一例大鼠死亡。结论:通过立体定向移植方法,移植干细胞入癫痫大鼠脑内CA1区,位置准确。
Objective: To explore the feasibility of stereotaxic trsnsplantation in stem cells Methods:The epileptic rat model were built by injected penicillin into peritoneal. The 24 Sprague -DawleySD healthy rats were divided into the normal saline transplanted group, and tern cells transplanted group. After 7 days the rats were perfused and the survival cells were observed by the fluorescenc microscope, at the same time make sure the accuracy of stem cell in transplanted area and safety of the stereotaxie transplantation. Results:After transplanted in stem cells, all the rats were survival, there were many cells with GFP around the pin hole. Conclusion:The bone marrow stem cell and neural stem cell were transplanted in CA1 in epilepic rats by stereotaxis -apparatus and the sites were accurate.
