硒蛋白在内皮细胞发育和心肌功能中的重要作用 被引量:6

摘要 应用LoxP-Cre技术消除内皮细胞、心肌细胞或骨骼肌细胞内的硒tRNA基因trsp,以明确硒蛋白在心血管疾病中的作用。内皮细胞的硒蛋白表达丧失可引起胚胎致死性的变化。14.5 d大的胚胎有中枢神经系统坏死,皮下出血和红细胞不成熟等发育异常。出生后第12天硒蛋白表达缺失的心肌细胞仍无明显的表型改变。硒蛋白表达缺失的小鼠表现出运动减少,呼吸过速直至死亡。病理学检查表明trsp缺失的小鼠会使中度或重度心肌炎扩展为纵隔炎。因此,硒蛋白表达缺失表明了硒蛋白在血管内皮细胞的发育和在心肌功能方面起重要作用。这些结果表明在这类细胞中硒蛋白的表达缺失和心血管疾病存在着直接联系。
