摘要
对四棱豆属栽培品种nrDNA ITS序列进行克隆测序,并进行序列比较与品种间亲缘关系分析.结果表明:(1)供试品种间ITS序列的变异小,GC含量低,为51.4%~51.6%.(2)供试四棱豆品种间遗传变异范围在0.0000~0.0009,表明四棱属内nrDNA ITS区存在一定的遗传变异,但变异较低,表明供试材料间亲缘关系较近.(3)在3种不同的分子聚类树中,K0021总位于不同的分支中,表明K0021与其他栽培品种属不同品系.Z8和M8、S3和M7在3种树中都同时聚为一类,表明彼此间亲缘关系很近.以红花菜豆为外类群进行聚类分析发现四棱豆属与其他豆科植物属间的亲缘关系较远.
Phylogenetic relationship of seven cultivars of Psophocarpus Neck. were esti- mated based on nrDNA ITS by using MEGA4.0 software in the study. The results showed: (1)The ITS sequences had a small variation intested materials, the content of GC was low,from 51.4% to 51.6%. (2) the range of genetic variation is from 0. 0000~0. 0009 among different Psophocarpus tetragonolobus,the sequence data revealed considerable variation, but the mutation rate was low, the seven cultivars of Psophocarpus Neck. had a near relationship, (3) In the three clustering tree K0021 showed belonging to different cultivars compaired with other cultivar. Z8 and M8 in the three trees were all clustered in the same branch, also S3 and MT, there was a close relationship between Z8 and M8, so was S3 and MT. The research showed the relationship was far between Psophocarpus tetragonolobus and Phaseolus coccineus L.. The study provided reference for classification of Psophocarpus Neck..
