摘要
目的:研究使用蛋清甘油作为支撑物的石蜡包埋制片技术用于基于细胞的形态学检测的可行性.方法:针对人胃癌BGC-823培养细胞、临床送检宫颈TCT检查细胞、胃癌标本残碎细胞液等细胞学材料,运用蛋清甘油作为支撑物进行石蜡包埋制片,进行HE染色及免疫细胞化学染色,并将染色效果与细胞爬片、细胞甩片及细胞涂片染色效果进行对照分析.结果:各种细胞标本在使用蛋清甘油作为支撑物的石蜡包埋制片技术进行处理后,HE染色见细胞分散均匀,染色清晰,细胞质及细胞膜境界清楚,免疫细胞化学染色见背景干净,阳性细胞明显,定位准确.两种染色效果均优于细胞爬片、细胞甩片及细胞涂片.结论:采用蛋清甘油作为支撑物的石蜡包埋制片技术处理各种细胞学标本,实验室条件要求低、操作方便且便于保存,用于常规HE染色以及免疫细胞化学染色取得了良好效果.
Objective:The feasibility of using protein glycerol as holdfast,paraffin imbedding and making slide technology to deal with different cell specimens.Methods:Protein glycerol as holdfast,paraffin imbedding and making slide technology were used to deal with human gastric cancer cell line BGC-823,cell from TCT examination of cervix of uterus and cell from gastric tissues.All slides of different cells were examined by HE and immunocytochemistry(ICC) methods.The staining effects were compared with the methods of climbing,throwing and smearing slides.Results:Using protein glycerol as holdfast,paraffin imbedding and making slide technology to deal with different cell specimens,HE staining was found that cell was well-distributed,staining was clear and boundary of membrane and nuclear was distinct,ICC staining was found that background was clear,positive cell was obvious and allocation of positive domain was accurate.The results were better than the methods of climbing,throwing and smearing slides.Conclusion:Using protein glycerol as holdfast,and paraffin imbedding technology to make slide,the requests of instruments are lower,performance is easier and preservation is more convenient than other methods.With cell paraffin imbedding method to do HE and immunocytochenmistry staining,the effect of staining is better than the methods of climbing,throwing and smearing slides.
