黄瓜扩张蛋白Cs-EXP10基因5'序列的克隆和表达的初步分析 被引量:1

摘要 扩张蛋白是促进植物细胞壁伸展的一类蛋白质,黄瓜果实膨大过程中扩张蛋白具有重要作用。本研究以发育中的黄瓜果实RNA为试验材料,通过RACE方法克隆了黄瓜Cs-EXP10基因的5'端未知序列,进而获得了该基因的全长,序列分析显示该基因编码287个氨基酸,有扩张蛋白的特征序列。Southern杂交表明Cs-EXP10在基因组中是以单拷贝形式存在的。其表达具有组织特异性,在膨大过程中的果实中表达量高,显示其与果实的膨大有关。在叶和茎组织中也有少量表达,而在根和花中基本不表达。 Expansin is a kind of protein to promote plant cell wall expanding,it is important for cucumber fruit enlargement during fruit development.Cucumber expansin 10(Cs-EXP10)gene 5-end sequence were clones by RACE method from the RNA of cucumber developing fruits,and then the total cDNA sequence of this gene were get.This gene coded 287 amino acids after sequence analysis.Southern blotting result showed that Cs-EXP10 was single copy in cucumber genome.Expression analysis showed that it expressed highly in expanding fruit,and express little in stems and leaves,however almost do not express in roots and flowers,which means it correlated with fruit expanding.
