摘要
目的观察启膈方对小鼠前胃癌移植瘤的上皮性钙黏附蛋白(E-Cad)、基质金属蛋白水解酶-2(MMP-2)、微血管密度(MVD)、血浆α颗粒膜蛋白(GMP-140)、血栓素B2(TXB2)的影响。方法小鼠前胃癌(FC)细胞接种在30只615小鼠右腋皮下,接种后随机分为启膈方组、氟尿嘧啶(5-FU)组、空白对照3组,每组各10只。接种后16 d摘除小鼠眼球取血,获得血浆备用,处死小鼠,剥离肿瘤组织。用链霉菌抗生物素蛋白-过氧化酶(SP)法检测各组肿瘤组织E-Cad、MMP-2、MVD的表达,用放射免疫法检测各组小鼠血浆TXB2水平,用酶联免疫法检测小鼠血浆GMP-140水平。结果启膈方组E-cad的表达强于空白对照组、5-FU组(P<0.01),5-FU组E-cad的表达强于空白对照组(P<0.05)。空白对照组MVD明显高于启膈方组(P<0.01)和5-FU组(P<0.05),启膈方组低于5-FU组(P<0.05)。启膈方组、5-FU组的MMP-2表达弱于空白对照组(P<0.05)、5-FU组与启膈方组比较差异无统计学意义(P>0.05)。启膈方组、5-FU组的血浆GMP-140与TXB2含量均明显低于空白对照组,尤其是血浆GMP-140含量,启膈方组、5-FU组与空白对照组比较差异有统计学意义(P<0.01),启膈方组与5-FU组之间差异无统计学意义(P>0.05)。血浆TXB2水平,启膈方组低于5-FU组,差异有统计学意义(P<0.05)。结论启膈方可通过提高E-Cad的表达,降低MMP-2的表达,降低MVD,降低小鼠血浆GMP-140、TXB2水平,起到抑制肿瘤转移的作用。
Objective To observe the effect of Qigefang on epithelial-cadherins(E-cad),matrix metalloproteinase-2(MMP-2),microvessel density(MVD),GMP-140,TXB2 of gastric cancer metastasis of mouse.Methods 30 mice of 615 were vaccinate by cell suspension of gastric cancer at right armpit hypo,a mouse 0.1 mL(1×106cell).Vaccinated mice were divided into three groups randomly,Qigefang group,5-FU group and control group.Each group was 10 mice.16 days of postinoculation,eye globe were removed to obtain blood.Then tumor architecture was stripped when mice were killed.To detect expression of E-Cad,MMP-2,MVD on tumor of each group by SP immunohistochemistry.To detect TXB2 level of mice blood plasma by radio-immunity.To detect GMP-140 level of mouse blood plasma by euzymelinked immunosorbent assay.Results Expression of E-cad of Qigefang group was stronger than that of control group and 5-FU group(P0.01).The 5-FU group was stronger than control group on the expression of E-cad(P0.05).MVD of control group was obviously higher than Qigefang and 5-FU group(P0.05).Qigefang group was lower than 5-FU group on MVD(P0.05).Expression of MMP-2 of Qigefang group was lower than control group(P0.05).Expression of MMP-2 of 5-FU group was also lower than control group(P0.05).Level of GMP-140 and TXB2 of mice blood plasma,Qigefang group and 5-FU group was lower than that of control group obviously,especially level of GMP-140 of blood plasma.There was significant difference between Qigefang group,5-FU group and control group(P0.01),but there was no significant difference between Qigefang group and 5-FU group(P0.05).Level of blood plasma TXB2 in Qigefang group was lower than that in 5-FU group(P0.05).Conclusion Qigefang can raise expression of adhesion molecule E-Cad,depress expression of MMP-2 and expression of MVD,and depress GMP-140,TXB2 level of mouse blood plasma.Therefore,Qigefang has inhibiting effect on metastasis.
