烤烟T-psy1基因结构及其内含子信息分析 被引量:1

摘要 八氢番茄红素合成酶(phytoene synthase)是调控类胡萝卜素合成和积累的关键酶之一,对烟叶质量和香气的形成起着重要作用。为获得烤烟八氢番茄红素合成酶基因的结构信息,利用GenBank数据库中烟草八氢番茄红素合成酶基因T-psy1基因cDNA序列(基因登录号为HM345582,蛋白登录号为ADK25054)信息,采用同源克隆法从烤烟品种K326叶片中克隆获得了T-psy1基因DNA序列。基因结构分析表明,T-psy1包含有6个外显子和5个内含子,外显子和内含子的剪接符合GT-AG原则,5个内含子的变化范围为96~504 bp。T-psy1基因结构及内含子信息为烟草T-psy1基因表达和烟草类胡萝卜素合成途径的调控提供了序列信息。 Phytoene synthase is one of the key enzymes which regulate the synthesis and accumulation of carotenoid and play roles in the formation of the cured-leaf quality and aroma.For the information of phytoene synthase gene structure of tobacco,the DNA sequence of T-psy1 gene was cloned from the leaves of flue-cured tobacco cultivars K326(Nicotiana tobacum cv.K326),based on the information of the cDNA sequence of T-psy1 gene cDNA sequence from the GenBank database(gene accession number:HM345582,protein accession number:ADK25054).Analysis of genomic structure showed that it contained 6 exons and 5 introns,and the splicing principles of its exons and intron were consistent with GT-AG,the size of 5 introns ranged from 96 bp to 504 bp.The genomic structure of T-psy1 and its introns provided the sequence information for study on the expression of T-psy1 and the regulation of tobacco carotenoid synthesis pathway.
