高效液相色谱法测定苦参片中苦参碱含量

Content Determination of Matrine in Kushen Tablets

目的建立测定苦参片中苦参碱含量的高效液相色谱法。方法色谱柱采用AlltechAlltimaAmino柱(250 mm×4.6 mm,5μm),流动相为乙腈-无水乙醇-3%磷酸溶液(80∶10∶10),流速1.0 mL/min,检测波长220 nm。结果苦参碱进样量在0.2064～3.048μg范围内与峰面积线性关系良好,回归方程Y=469.76X-3.2632(r=0.999 8),平均回收率为97.99%,RSD=1.21%(n=6)。结论所建立的高效液相色谱法简便、快速、重复性好,可用于苦参片的质量控制。

Objective To establish a method to determine the contents of matrine in Kushen Tablets.Methods An HPLC method was adopted. Alltech Alltima Amino column（250 mm×4.6 mm,5μm）was used.The mobile phase consisted of acetonitrile-anhydrous alcohol-3% phosphoric acid solution（80∶10∶10）at a flow rate of 1.0 mL/min;the wavelength of detector was 220 nm.Results The calibration curves of matrine showed good linearity in the range of 0.206 4-3.048μg.The regression equation was Y=469.76 X-3.263 2（r=0.999 8）.The average recovery rate was 97.99%,RSD=1.21%（n=6）.Conclusion This method is simple,high-speed and reproducible,and is suitable for the determination of matrine contents in Kushen Tablets.
