摘要
目的:探讨PLGA神经导管内移植雪旺氏细胞(SCs)对猫颅内段动眼神经再生的影响。方法:将22只家猫随机分为3组,导管组10只,SCs组10只,对照组2只。右侧动眼神经制作4 mm缺损后,分别用PLGA导管、PLGA导管+SCs悬液修复,对照组不修复。14周时处死动物,取远端再生神经,用光镜、电镜观察及轴突图像分析评估修复效果。结果:术后14周时,导管组有7只猫,SCs组有8只猫右侧神经功能基本恢复,神经连续性恢复;对照组无恢复。导管组及SCs组再生神经有髓纤维的数目分别为(8 760±597)(、10 255±1 271),导管组及SCs组再生神经平均轴突直径为(4.16±0.30)um、(4.59±0.34)um;两组间纤维数目、轴突直径差异有统计学意义(P<0.05)。结论:PLGA导管内移植同种异体雪旺氏细胞悬液能促进猫颅内段动眼神经再生。
Objective: To evaluate the effects of nerve regeneration when the oculomotor nerve defects were repaired with PLGA conduit and Schwann cells(SCs) in cat models.Methods: A total of 22 cats were randomly divvied into conduit group(n=10),SCs group(n=10),and control group(n=2).The cistern segment of the right oculomotor nerve was transected to make a 4 mm nerve defect in each cat.Then,nerve defects in the conduit group were repaired with PLGA nerve conduits,in SCs group with PLGA conduit and SCs solution, while no repairing were conducted for the control group. Cats were sacrificed 14 weeks after the surgery, never regenerating status were observed and evaluated by microscope and electron microscope. Results: 14 weeks after surgery, there were 7 cats in the conduit group and 8 cats in the SCs group showed recovery of oculomotor nerve function and anatomical continuity, while no improvement were found in the control groups. Number of mean nerve fiber of the distal stump were (8 760~597)in conduit group, and stump were (4.16--+_0. (10 255-+-1 271) 30) /~m in conduit in SCs group, respectively; mean axonal diameter of the distal group, and (4.59~0.34) /,m in SCs group, both showing significant difference between the two groups (P both d0.05). Conclusions: Local implantation of SCs solution in PLGA conduit can promote the oculomotor nerve regeneration in cats.
