摘要
在单因素试验的基础上,采用响应面分析法对高产淀粉酶菌株的培养条件进行了优化.通过单因素试验,评价了不同温度、pH值、时间、碳源浓度、氮源浓度和摇床转速等6个因素对菌株产淀粉酶含量的影响.对蛋白胨质量分数、玉米粉质量分数、pH值和温度等主要影响因素进行中心组合设计,响应面分析结果为pH值对产酶影响显著,最佳培养条件为pH值为6.5,玉米粉质量分数为0.3%、蛋白胨质量分数为0.5%、培养温度为35℃、培养时间为30 h.
In the single-factor test,high-yielding strains of amylase culture conditions were optimized based on the response surface analysis method.Through single factor test,the effects on produced amylase of the different temperature,pH,time,carbon source concentration,nitrogen concentration,and shaker speed were concluded.After the central composite design of peptone concentration,the concentration of corn flour,pH and temperature and other major factors,got the result from the response surface analysis that the pH effect on enzyme production significantly,the best culture conditions for pH=6.5,the concentration of 0.3% corn meal,the concentration of peptone 0.5%,incubation temperature of 35 ℃,the culture time of 30 hours.
