摘要
应用抑制消减杂交技术(Suppression subtractive hybridization,SSH)分离‘安吉白茶’全白期与全绿期叶片差异表达基因,成功构建了‘安吉白茶’白期叶与绿期叶的正、反向消减文库。随机挑选80个阳性克隆测序,通过BLASTX比对分析,80个序列中有30个没有找到任何同源序列,8个序列太短,且同源性不高,42个序列与已知蛋白有着或高或低的同源性。其中有3个基因片段可能与‘安吉白茶’高氨基酸形成相关。
To separate the differentially expressed genes between white and green period leaf of'Anji Baicha',a subtracted library was constructed by the suppression subtractive hybridization(SSH) method.By sequencing the 80 recombinant clones and through BlastX analysis,30 showed no homologous,8 had short sequence and also showed poor homologous,for the other 42 fragments in 80 recombinant clones.They showed more or less homologous to all proteins in the database.3 fragments might have correlation with high content of amino acidin‘Anji Baicha’.
