摘要
研究旨在脂质体介导基因转染巴马猪胎儿成纤维细胞时获得较高转染效率,同时以GFP为报告基因探讨影响外缘基因转染效率的参数,如DNA和脂质体的用量、转染的细胞数量及细胞与DNA与脂质体复合物的孵育时间。结果表明,6μgDNA与15μL脂质体转染8h,可获得较满意的转染效率。
This study was aimed to obtain higher efficiency in gene transfection into Bama Pig Fetal Fibroblasts and to study the role of green fluorescence protein (GFP) as a reporter system.We investigated the effect of parameter such as the dose of DNA and liposome number of cell transfected and exposure time of the cell to the DNA-liposome complexes.It was indicated that 6 μg DNA with 15 μL liposome resulted in the highest transfection efficiency after 8 h transfection.
