归延胶囊对脑缺血再灌注时相点大鼠皮层神经细胞凋亡的影响 被引量:1

Effect of Guiyan Capsule on neural cell apoptosis of cerebral cortex of cerebral ischemia reperfusion at different times rats

摘要 目的:观察归延胶囊(当归、莪术和延胡索)对脑缺血再灌注(CI/R)时相点大鼠皮层神经细胞凋亡的影响,探讨治疗CI/R损伤的作用机制。方法:采用大鼠脑中动脉阻断制备局灶性CI/R模型,缺血2 h再灌注6 h、24 h、48 h、3 d、10d、30 d,取脑组织,干湿重量法测定脑含水量,TTC染色检测梗死面积,TUNEL检测神经细胞凋亡,RT-PCR检测损伤侧脑皮层组织Fas、Bax、Caspase mRNA表达,Western Blot检测损伤侧脑组织Fas、Fas-l、Bax、BCl-2、Caspase-3、9蛋白表达。结果:归延能有效地改善CI/R所致的脑皮层病理组织学损伤,降低各时相点脑梗死面积,降低再灌注48 h、3 d、10 d脑含水量,减少再灌注48 h、3 d、10 d、30 d脑皮层神经细胞凋亡数,下调各时相点损伤侧Fas、Bax、Caspase-3、9 mRNA及蛋白表达。结论:归延抑制脑皮层神经细胞凋亡的作用机制可能是通过调节Bcl-2/Bax、Fas/FasL和Caspase-3的活性而实现的, AIM:To observe the effect of Guiyan Capsule(Angelicae sinensis Radix,Curcumae Rhizoma,Co-rydalis Rhizoma) on neural apoptosis in injury of cerebral ischemia reperfusion.METHODS: Focal cerebral ische-mia reperfusion in rats was made by transient occlusion of middle artery for 2 h,then reperfused in 6 h,24 h,48 h,3 d,10 d,30 d.The cerebral edema was measured with dry-wet weight.The infarct size was determained with 2,3,5-triphenlytetrazolium chloride stain;the neural cell apoptosis was detected by TUNEL.After reperfusion for 6 h,24 h 48 h,3 d,10 d,the expression of Fas,Bax,Caspase mRNA in cerebral cortex were determined by RT-PCR;the protein expression of Fas,Fas-l,Bax,BCl-2,Caspase-3、9 in cerebral cortex were detected by western blod-ing.RESULTS: Guiyan Capsule improved the pathohistologic lesion in cerebral ischemia reperfusion,reduced the infarct size at different times and cerebral edema content during reperfusion in 48 h,3 d,10 d;decreased the neural cll apoptosis,down-regulated the expression of Fas,Bax,Caspase-3、9mRNA and protein in cerebral cortex at dif-ferent times.CONCLUSION: These results show the inhibitory effect of Guiyan Capsule on neural cell apoptosi in cerebral cortex may be correlated with regulating the activity of Bcl-2 /Bax,Fas/FasL and Caspase-3.
