薏苡仁注射液(康莱特)联合顺铂对人肺腺癌细胞A549抑制作用及机制 被引量:20

摘要 目的:探讨薏苡仁注射液(康莱特)联合化疗药物顺铂(CDDP)对人肺腺癌细胞A549抑制作用的机制。方法:体外培养人肺腺癌细胞系A549并以康莱特、顺铂进行处理,SRB法检测药物对各组细胞的抑制率;逆转录-聚合酶链反应法检测多药耐药因子MDR1、Bcl-2和Bax mRNA在各组中表达情况;Western blot法检测P-gp、Bcl-2、Bax蛋白水平表达;同时检测康莱特对正常细胞的作用。将各组结果进行比较。结果:康莱特组A549细胞存活率较对照组明显降低,康莱特联合顺铂导致细胞存活率进一步下降(均P<0.05),而康莱特对正常细胞无明显抑制作用(P>0.05);康莱特导致A549细胞MDR1和Bcl-2 mRNA水平、蛋白水平均较空白对照组降低(均P<0.05),而Bax mRNA和蛋白水平则升高(均P<0.05);康莱特与顺铂联合作用于A549细胞时这一变化更为显著(均P<0.01);康莱特对正常细胞系的耐药因子则无明显作用(均P>0.05)。结论:康莱特可抑制肺腺癌细胞生长,该药具有明显的逆转肺腺癌多药耐药性的作用,与化疗药物联合使用可提高肿瘤细胞对化疗药物的敏感性。 AIM:To investigate the effect and mechanism of Kanglaite Injection associated with Cisplatin(CD-DP) in lung adno carcinoma cells A549.METHODS:The lung adno carcinoma cells A549 was treated by Kang-laite Injection associated with CDDP in different groups,then inhibition rates were measured by SRB assay in vitro.The expressions of MDR1/P-gp,Bcl-2,and Bax were determined by reverse transcription-polymerase chain reaction and Western blot in A549 cells.The effect of Kanglaite on human brain microvascular endothelial cells(HB-MEC) was also tested.RESULTS:The survival rate of A549 cells treated by Kanglaite was lower than that of the control group(all P0.05),and the survival rate decreased more when Kanglaite combined with CDDP;Kang-laite Injection had no obvious inhibition on HBMEC.The levels of MDR1/P-gp,Bcl-2(mRNA and proteins)were lower in cells after being treated by Kanglaite(all P0.05),but expression of Bax was higher(P0.05);also Kanglaite had no obvious influence on these factors in HBMEC(all P0.05).CONCLUSION:Kanglaite can in-hibit the growth of A549 cells,and it also can reverse multidrug resistance of A549 by regulating the expression of multidrug resistance associated factors.Combination of Kanglaite and CDDP can increase chemosensitivity of A549 cells.
