摘要
目的:通过配伍中药白芍考察对川楝子的减毒效应及其机制,以探求减轻川楝子肝毒性的中药复方减毒配伍规律。方法:SD大鼠,分组,单次灌服蒸馏水或川楝子、川楝子+白芍,放射免疫法检测肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)含量、免疫组化法检测核转录因子-κB(NF-κB)、ICAM-1蛋白表达。Western-blot法检测caspase-3、bcl-2基因表达。结果:白芍与川楝子配伍应用后,能够对抗肝组织TNF-α、IL-6水平的提高(P<0.05或P<0.01),能对抗肝组织NF-κB、ICAM-1蛋白表达的增强(P<0.01);能够调节肝组织caspase-3、bcl-2的基因表达。结论:白芍能对抗川楝子导致的肝损伤,其减毒机制是该药可以减轻肝组织炎症反应,并与调节肝细胞坏死相关基因caspase-3、Bcl-2的表达有关。
AIM:To find the way to reduce the hepatic toxicity of Fructus Toosendan by the use of Radix Paeoniae Alba METHODS:SD rats were assigned to three experimental groups:control group(given distilled water),Fructus Toosendan group and Fructus Toosendan + Radix Paeoniae Alba group.The contents of tumor necrosis factor-alpha(TNF-α) and interleukin-6(IL-6) were determined by ELISA;The protein expression of nuclear transcription factor-κB(NF-κB) and intercellular adhesion molecule-1(ICAM-1) were detected by immunohistochemistry;The expression of caspase-3 and bcl-2 were detected by Western-blot.RESULTS:After adding Radix Paeoniae Alba to Fructus Toosendan,it could antagonize the enhancement of TNF-α and IL-6 level in the rats'hepatic tissue and the protein expression of NF-κB and ICAM-1 significantly(P0.05 or P0.01),also could be againstthe enhancement of the genetic expression of caspase-3 and the decrease of the genetic expression of bcl-2.CONCLUSION:Radix Paeoniae Alba can lower the hepatic injury induced by Fructus Toosendan via lessening inflam-matory reaction in relation to caspase-3 and bcl-2.
