非角化性鼻咽癌组织中血管内皮细胞粘附因子mRNA的检测 被引量:1

The detection and clinical signicance of VCAM-1 mRNA in unkeratinized nasopharyngeal carcinoma tissues

摘要 目的探讨VCAM-1 mRNA增加表达与浸润淋巴细胞和临床分期、淋巴结转移的关系。方法收集34例非角化性鼻咽癌病例的临床资料和活检组织,用原位杂交方法检测鼻咽癌组织中的VCAM-1 mRNA,同时在HE切片上观察淋巴细胞浸润情况。结果 VCAM-1mRNA在97.11%(33/34)的非角化性鼻咽癌癌细胞中被检测到。其积聚强度与鼻咽癌临床分期和有无淋巴结转移无明显相关(P=0.098,P>0.05)及与癌巢间和癌巢内淋巴细胞浸润程度也无关(P=0.092,P=0.479)。21例可见鼻咽被覆上皮组织,均被检测到VCAM-1 mRNA,其中12例有部分血管内皮细胞也被检测到VCAM-1 mRNA。结论 VCAM-1mRNA积聚程度与鼻咽癌组织中淋巴细胞浸润程度和肿瘤进展无明显相关。血管内皮细胞不是鼻咽癌组织中VCAM-1mRNA积聚的主要细胞,在鼻咽癌组织中多种细胞中被检测到,提示鼻咽癌组织中存在着较强的诱导VCAM-1转录的因素。 Aim To invesrtigate the signicance of VCAM-1 mRNA in unkeratinized nasopharyngeal carcinoma tissues (UNPC). Methods VCAM-1 mRNA was assessed in paraffin embedded UNPC specimens (n=34) by in situ hybridization,meanwhile the lymphocytes infiltrating in the UNPC tissue were observed on HE specimens and clinical data were collected. Results The positive signal of VCAM-1 mRNA was observed in 97.11% (33/34) UNPC cases without correlation with either tumor staging (P=0.098),lymph node metastasis (PM}.05), or the intension of lymphocytes infiltration in inter- or intra-carcinoma nest(P=0.092,P=0.479). The signal of VCAM-1 mRNA was seen on all nasopharynge.al mucosa epithelia (n=21),but only part of vascular endothelial cell of capillary or venule in a third of the cases (12/34,37.5%). Conclusion High level transcription of VCAM-1 mRNA in UNPC is a mason of high expression of VCAM-1 protein in NPC cells. No significant correlation between quantity of VCAM-1 mRNA and lymphocytes inf'dtration or tumor staging; Vascular endothelia in UNPC is not the pominant site where the VCAM-1 mRNA transcription increases; Several kinds of cells showing VCAM-1 mRNA positive signal imply that some effective factors inducing VCAM-1 expression might exist in UNPC tissue.
