NaCl、PEG双重胁迫下转DREB1A基因多年生黑麦草T1代萌发期抗性研究 被引量:3

A Study on Tolerance of T1 Generation of Transgenic Perennial Ryegrass with DREB1A Gene at Germination Period under PEG and NaCl Double Stress

摘要 对转DREB1A基因多年生黑麦草(Lolium perenne)T1代种子和多年生黑麦草非转化植株F1代种子分别用1.5%NaCl溶液、15%PEG溶液、1.5%NaCl+15%PEG进行处理。测定了相对发芽势、相对发芽率、胚芽长、胚根长、细胞膜透性、相对含水量、叶绿素含量、萎蔫幼苗数、存活率等指标,研究了供试材料对单独胁迫和双重胁迫的抗性,并进行对比。相对发芽势、相对发芽率、胚芽长、细胞膜透性、叶绿素含量等指标的测定结果显示,DREB1A对三种胁迫的抗性强度依次为NaCl>PEG>NaCl+PEG。叶片相对含水量、存活率等指标的测定结果表明DREB1A对三种胁迫的抗性强度依次为PEG>NaCl>NaCl+PEG。萎蔫幼苗株数的测定结果显示,DREB1A对三种胁迫的抗性强度依次为NaCl>NaCl+PEG>PEG。胚根长在三种胁迫下的响应差异不显著。DREB1A对NaCl、PEG单独胁迫和NaCl+PEG双重胁迫均具有一定程度抗性,比非转化植株的抗性明显增强。转DREB1A基因多年生黑麦草T1代种子可以作为干旱、盐碱地区改良和利用的植物资源。 In order to understand the tolerance of T1 generation of transgenic perennial ryegrass(Lolium perenne) with DREB1A gene under double stress,the research was focused on the analysis of the physiological properties of transgenic plants after PEG,NaCl and PEG+ NaCl treatment at germination stage,with the control group of water treatment.The result of relative germination potential,relative germination rate,seedling height,relative conductivity,wilting degree and chlorophyll content showed that all materials with DREB1A gene showed the highest tolerance under NaCl stress.The results of relative water content and survival rate indicated that materials with DREB1A gene had the highest tolerance under PEG stress.All materials with DREB1A gene had significant tolerance under PEG+ NaCl stress and the difference between transgenic plants and CK was significant.This suggested that T1 generation of transgenic perennial ryegrass was stress-tolerant and could be extended in drought and salinity soil to improve drought and salinity land.
