摘要
文章通过RT-PCR技术克隆了牦牛PRDM9基因的cDNA序列,利用DNAMAN、MAGA4、ExPASy等多个生物信息学软件进行了分析研究。结果表明:扩增出的牦牛PRDM9基因的编码区长1 533 bp,编码510个氨基酸。与普通牛、人和鼠相应基因核苷酸序列进行比对,其一致性分别为99%、76%和69%。预测的牦牛PRDM9蛋白二级和三级结构显示它是一个具有3种功能结构域的弱碱性螺旋状蛋白。这为今后的进一步研究提供了理论基础。
The objectives of this study were to clone the coding region of PRDM9 gene in order to provide a theoretical basis for the study of fertilization biology in the yak by PT-PCR.The nucleotide and amino acid sequences were analyzed using DNAMAN,MAGA4 and ExPASy software.Results showed that the coding region of PRDM9 gene in yak was 1 533 bp long and encoded 510 amino acids.Compared with Bos taurus,human and mouse,the similarities of PRDM9 gene sequence in yak were 99%,76%,69%,respectirely.The coding region of PRDM9 gene in yak was successfully cloned,which provides a theoretical basis for further study on its structure and function.
