奶牛CRP基因多态性与奶牛乳腺炎及乳质乳量的相关性研究及生物信息学分析

Polymorphism of CRP gene and its relationship with mastitis and milk in Chinese Holstein and bioinformatics analysis

摘要 选用400头中国荷斯坦奶牛取血样400份,每份各2管。根据奶牛CRP基因序列设计2对引物,利用PCR-SSCP技术检测CRP基因的多态性,并用透射免疫比浊法测定奶牛血清CRP水平,然后运用SPSS等统计学软件对其与乳房炎及乳质、乳量的关系进行统计分析。结果显示,在第1对引物的扩增片段中检测到多态位点,出现3种基因型(AA、AB、BB),第2对引物的扩增片段中并未检测到多态位点。将第1对引物所扩片段的不同基因型与奶牛乳腺炎的相关性作关联分析,结果3种不同基因型奶牛的体细胞数、乳质率、蛋白率、305d产奶量均无显著差异,但患乳房炎奶牛个体全为AA型。而CRP含量检测结果可知患乳房炎的奶牛与正常个体之间的血清CRP含量并无显著差异。 The goal of the present study was to detect the polymorphism in C-reactive protein(CRP) and analyze the relationship between CRP gene and mastitis(include resistance and susceptible to mastitis,milk quantity and quality),and the level of serum CRP between mastitis and normal cow.According to the sequence of bovine CRP gene,design two pairs of primers and use PCR-SSCP to detect single nucleotide polymorphism of CRP gene in Chinese Holstein,and analyze the relationship through the software SPSS and Excel.There was one polymorphic locus occurring in P1 with three genotypes(AA,AB,BB),and no polymorphic locus occurring in P2.The relationship analysis indicates that the genotype and the mastitis have no significant difference,and also do to the milk quality and quantity,but all of the mastitis dairy cows are AA genotype.And the serum CRP level between mastitis and normal group are not significantly different.
