摘要
目的研究针对CD147 RNA干扰对前列腺癌PC-3细胞系生物学行为的影响。方法利用针对CD147 RNA干扰片段构建的表达载体转染PC-3细胞,获得稳定表达株。通过RT-PCR和Western blotting鉴定后进行前列腺癌细胞侵袭力实验和动物实验。结果 RT-PCR和Western blotting结果表明干扰片段可以较好地封闭前列腺癌PC-3细胞CD147分子mRNA和蛋白表达水平;与转染空质粒组相比,CD147干扰组前列腺癌PC-3细胞侵袭力下降,荷瘤裸鼠成瘤力下降(P<0.05),抑瘤率达32.82%。结论 RNA干扰封闭CD147基因能抑制前列腺癌细胞的侵袭力和成瘤力,有可能成为前列腺癌基因治疗的新途径。
【Objective】 To study the biological effects of targeting CD147 RNA interference(RNAi) on prostate cancer PC-3 cell line.【Methods】 PC-3 cells were transfected with a vector coding for a pairs of small interfering RNAs(siRNA) targeting CD147.Then,we obtained the stably expression PC-3 cell lines.RT-PCR and Western blotting were employed to determine the effect on expression of CD147 mRNA and protein.Matrigel invasion assay was performed to evaluate the invasion ability of PC-3 cells in vitro.Cells were transplanted subcutaneously in nude mice to observe the tumorigenesis.【Results】 Our results showed that CD147 expression was significantly inhibited by siRNA transfectants in PC-3 cells at mRNA and protein levels.Invasion abilities in tumor cells and tumorigenesis in nude mice were lowered in the experiment group than those in the control group(P 0.05).Tumorigenicity inhibition rate reached 32.82% in the experimental group.【Conclusions】 CD147 downregulation via RNA interference technology could decrease the invasive capability and tumorigenesis of prostate cancer cells.It was demonstrated that targeting CD147 could potentially be a new approach for prostate cancer gene therapy.
