摘要
目的:研究复方丹参方中使药降香不同成分对君药丹参的主要成分丹参素药动学的影响。方法:给家兔灌服丹参水提物、丹参-降香水提物和丹参-降香油,采用HPLC法检测不同采样点血浆中丹参素的血药浓度,用DAS2.0数据处理软件计算药动学参数。结果:配伍前后,药-时曲线均呈二室开放模型,与丹参水提物组比较,配伍组曲线下面积AUC0-∞均明显增加,清除率(CI/F)减小,丹参-降香油组Cmax明显增加,t1/2a和tmax明显延长。结论:使药降香水提物和降香油均可提高君药丹参有效成分丹参素体内生物利用度,减少丹参素在体内消除,降香油为使药降香引药归经的主要活性成分。
OBJECTIVE To study the effect of different components of "Shi herb" Dalbergia odoriJera (IX)) on pharmaco kinetics of Danshensu(DSS), the main water soluble component of "Jun herb" Salvia miltiorrhiza (SM), in Danshen compound recipes in rabbits. METHODS Rabbits were orally administrated with SM decoction, SM coupled with DO aqueous ex tract, and SM coupled with DO volatile oil respectively. The concentration of DSS in plasma was determined at different sampling points by reversed phase high performance liquid chromatography, and the pharmacokinetic parameters were calculated by the software of DAS 2.0. RESULTS Before and after co administration, the time data of DSS concentration were all fitted with the two-compartment open model. Compared with SM decoction single administrated, the area under the plasma drug concentration time curves (AUC ) of the other two groups increased remarkably, the clearance rate(CU/F)decreased. Cmax of the SM coupled with DO volatile oil group increased remarkably, and t, 2,,and t were significantly prolonged. CONCLUSION Both aqueous extract and volatile oil of DO can increase the bioavailability of Danshensu, the active component of the "Jun herh" SM, and can reduce the elimination of Danshensu in uivo. DO volatile oil is the major active component of thc "'Shi herb" DO.
