摘要
目的评价负载8×10-5 mol/L银离子后珊瑚羟基磷灰石(CHA)的生物相容性。方法采用珊瑚羟基磷灰石材料与AgNO3及NaCl复合,经真空冷冻干燥后制备成8×10-5 mol/L Ag+浓度的载银珊瑚羟基磷灰石(Ag-CHA),并对健康成年昆明小白鼠、新西兰大白兔、白化豚鼠进行了全身急性毒性实验、热原实验、溶血实验和致敏实验。结果全身急性毒性实验中,负载8×10-5 mol/L银离子的Ag-CHA材料对小鼠无全身急性毒性作用,72 h内小鼠无毒性表现,体重无下降;热原实验中,家兔体温升高数最高分别为0.2、0.2、0.4℃,材料无热原反应;材料的急性溶血率为0.64%,无溶血作用;致敏实验中,实验组豚鼠皮肤无红斑无水肿。结论载银浓度为8×10-5 mol/L的Ag-CHA有良好的生物相容性,无全身急性毒性作用和溶血作用,无热原和致敏原。
Objective To evaluate the biocompatibility of silver-loaded coral hydroxyapatite(Ag-CHA) at 8×10-5 mol/L Ag+ concentration.Methods Silver-loaded coral hydroxyapatite(Ag-CHA) at 8×10-5 mol/L Ag+ concentration was prepared by treating with AgNO3 and NaCl solution,followed by vacuum freeze-drying.The biocompatibility of Ag-CHA was assessed with systemic acute toxicity test,pyrogen test,hemolysis test and dermal sensitization test on healthy adult Kunming mice,New Zealand rabbits,and albino guinea pigs,respectively.Results In systemic acute toxicity test,no significant weight lost or toxic reaction was observed in 72 hours with 8×10-5 mol/L Ag-CHA had no systemic acute toxicity.In pyrogen test,the highest temperature increments of rabbits were 0.2,0.2 and 0.4℃,respectively.In hemolysis test,the hemolysis rate was 0.64%.In dermal sensitization test,no erythema or hydroderma was revealed.Conclusion Ag-CHA at 8×10-5 mol/L Ag+ concentration provides good biocompatibility without systemic acute toxicity,pyrogenic effect,hematolysis or allergen.
