摘要
目的探讨钙调蛋白抑制剂三氟拉嗪(TFP)对戊四氮所致慢性癫痫大鼠模型癫痫发作及学习和记忆的影响。方法 40只大鼠随机分为4组,空白对照组、戊四氮(PTZ)诱导癫痫慢性模型组及三氟拉嗪高、低剂量组,观察大鼠行为学改变,应用Morris水迷宫试验,检测大鼠学习和记忆能力。结果模型组大鼠均有发作,其中2只5级发作,而TFP组发作程度减轻,以高剂量组为明显。Morris水迷宫试验:定位航行试验:TFP组大鼠在2 min内找到平台,时间与对照组比较明显缩短(P<0.05),以高剂量组明显;空间探索试验:与对照组比较,TFP组第一象限停留时间延长(P<0.05),穿环指数增加(P<0.05)。结论 TFP能减轻PTZ致痫大鼠发作程度,提高大鼠的学习和记忆能力。
Objective To observe the effects of trifluoperazine(TFP) on the learning ability and memory on rat models of Pentylentetrazol-induced epilepsy.Methods Forty rats were randomly divided into 4 groups,including sham group,control group,low TFP group and high TFP group.Behaviors of rats were observed according to the criteria of Racine.Meanwhile,the learning ability and memory were evaluated by Morris water maze test.Results Severe epilepsy was observed in control group,with two suffered of 5 grade seizure.TFP obviously reduced epilepsy in TFP groups in a dose-dependent manner.Total escape latency was significantly reduced in TFP groups in the place navigation trial(P0.05).Meanwhile,significantly prolong of first platform quadrant latency and platform crossing index were observed in TFP groups in spatial probe trial(P0.05).Conclusion TFP reduces PTZ-induced seizures and increase the learning ability of memory in rats.
