摘要
目的研究不同分子量的罗非鱼鱼鳞胶原蛋白多肽对体外培养新生小鼠颅骨MC3T3-E1细胞增殖与Ⅰ型胶原蛋白分泌及Ⅰ型胶原蛋白mRNA表达的影响。方法采用WST-8法测定胶原蛋白多肽对小鼠成骨细胞株MC3T3-E1细胞增殖的影响,ELISA法检测胶原蛋白多肽对MC3T3-E1细胞Ⅰ型胶原蛋白分泌的影响,采用real-time PCR分析不同分子量的胶原蛋白多肽对Ⅰ型胶原蛋白mRNA表达的影响。结果胶原蛋白多肽能促进MC3T3-E1细胞增殖(P<0.01)、促进MC3T3-E1细胞Ⅰ型胶原蛋白的分泌和Ⅰ型胶原蛋白mRNA的表达(P<0.05,P<0.01)。结论罗非鱼鱼鳞胶原蛋白多肽能有效促进MC3T3-E1细胞增殖与Ⅰ型胶原蛋白分泌,分子量较小的多肽(917.3Da)作用更加明显。
OBJECTIVE To study the effects of different molecular weights of collagen polypeptides from tilapia fish scale on proliferation of mouse osteoblastic MC3T3-E1 cell,type Ⅰ collagen secretion and mRNA expression in MC3T3-E1 cell.METHODS Cell proliferation viability was detected by WST-8 assay.ELISA was used to detect the secretion of typeⅠcollagen of cultured osteoblast MC3T3-E1 cell,and the expression of typeⅠcollagen mRNA in MC3T3-E1 cell was detected by real-time PCR.RESULTS The results of the experiment showed that the collagen peptide could stimulate the mouse osteoblasts proliferation(P0.01),and could increase typeⅠcollagen secretion and expression of typeⅠcollagen mRNA on MC3T3-E1 cell(P0.05,P0.01).CONCLUSION Collagen polypeptides from tilapia fish scale can stimulate MC3T3-E1 cell proliferation,and the latter can meanwhile increase typeⅠcollagen secretion.
