反式白藜芦醇对Aβ_(25-35)损伤大鼠海马旁回caspase-8、caspase-9的影响 被引量:4

摘要 目的观察反式白藜芦醇(TR)对β-淀粉样肽(Aβ25-35)损伤大鼠的海马旁回的作用。方法大鼠随机分为假手术组、模型组、TR高、低剂量组、多奈哌齐对照组。在大鼠海马内注射Aβ25-35后分别灌胃给相应药物10 d,采用TUNEL法检测5组大鼠海马细胞及周围皮质凋亡情况;采用real time RT-PCR法检测5组大鼠海马及皮质caspase-8、caspase-9 mRNA表达。结果大鼠海马及周边脑组织细胞凋亡情况,模型组与其他各组差异显著(P<0.01),说明造模成功。TR对caspases-8、caspase-9表达水平的下调非常显著(P<0.01),低剂量组与高剂量组之间也有统计学差异(P<0.05)。结论 TR对损伤的大鼠海马周围皮层神经元有保护作用,其机制可能部分与下调caspase-8、caspase-9 mRNA表达有关。 Purpose To observe the effects of ingredient of polygonum cuspidatum trans-resveratrol(TR) on injured rats parahippocampal gyrus.Methods Rats were randomly divided into 5 groups:the sham group,model group,TR high,low dose group and Aricept(Donepezil Hydrochloride Tablets) control group.In the rat hippocampus after injection of Aβ25-35,the drugs was administrated 10 days,5 groups assay and the surrounding cortex hippocampal cell apoptosis by TUNEL,using real time RT-PCR detected 5 rat hippocampus and cortex expression of the caspase-8,caspase-9 mRNA.Results The rat hippocampus and surrounding brain tissue apoptosis of model group was different to other groups(P0.01).The model was developed.The expression level of TR on caspases-8,caspase-9 were different to other groups(P0.01).There was also different between TR high and low dose group(P0.05).Conclusion hippocampus has a protective effect on injured rats parahippocampal gyrus,which may be partly reduced caspase-8,caspase-9 mRNA expression.
