摘要
[目的]研究软籽石榴离体叶片愈伤组织诱导的最佳条件。[方法]以软籽石榴的幼嫩叶片为外植体,以MS为基本培养基,研究了不同浓度和类型的植物生长调节剂、叶片的不同部分、不同糖浓度和类型对愈伤组织诱导的影响。[结果]培养基MS+0.1 mg/LNAA+0.1 mg/L IBA+1.0 mg/L 6-BA+30 g/L蔗糖对愈伤组织的诱导效果最好,选择叶片中部或叶片基部为外植体,可以有效提高诱导率,降低褐化率,诱导率最高达93.3%,褐化率最低达23.3%。[结论]为石榴的离体快繁、种质保存和利用基因工程技术改良品质奠定一定的理论和实践基础。
[Objective] The aim was to study the best conditions of callus induction conditions of vitro leaves from soft-seed Punica granatum L.[Method] The vitro leaves of soft-seed Punica granatum L.was as explant,MS was basic medium.Effect of different concentrations and types of hormone,different parts of the leaves and different sugar concentration and types on callus induction were studyed.[Result]The results showed that the medium MS+0.1 mg/L/NAA+0.1 mg/L IBA+1.0 mg/L 6-BA+30 g/L sucrose was best for callus induction,the base or middle of leaves(including part of petiole) was as explant,lt could improve callus rate and reduce browning rate.[Conclusion]The study provided theory and practice basic for callus induction,germinal conversation and improving quality.of soft-seed Punica granatum L.
