摘要
目的优化水相反应体系中的共轭亚油酸与薄荷醇的酶催化合成反应条件。方法根据中心组合试验设计原理,采用五因素五水平的响应曲面分析法,以反应转化率(%)为响应值作响应面和等高线图,考察了温度、反应时间、酶量、底物物质的量比、水量的影响。结果优化后的酶催化反应条件为:温度35℃、反应时间36 h、酶量40%、底物物质的量比为1,水量30%。结论本文所建立的数学模型显著(P<0.05),可用来对水相体系中的共轭亚油酸-薄荷醇酯的酶催化合成反应进行分析和预测。
OBJECTIVE To optimize the reaction condition of lipase-catalyzed synthesis of conjugated linoleic acid(CLA) with L-menthyl in aqueous medium.METHODS Response surface analysis(RSM) with 5 factors and 5 levels was employed according to central composite rotatable design.Response surface and contour were graphed with reaction conversion(%) as the response value.The effects of temperature,reaction time,enzyme amount,water content,and molar ratio were examined.RESULTS The optimal lipase-catalyzed reaction conditions were: temperature 35 ℃,reaction time 36 h,enzyme amount 40%,molar ratio 1,water content 30%.CONCLUSION The established mathematical model is was significant(P0.05),and may be applied to the analysis and prediction of the lipase-catalyzed synthesis of CLA with L-menthyl in aqueous medium.
