摘要
目的探讨侧脑室注射P物质(SP)在电针治疗局灶性脑缺血过程中的作用。方法将50只造模成功Wistar大鼠随机分为模型组、生理盐水组、SP组、电针组和SP+电针组,另设健康对照组,每组各10只。采用大脑中动脉阻塞(MCAO)复制大鼠局灶性脑缺血模型。生理盐水组和SP组大鼠分别在MCAO造模成功后侧脑室注射生理盐水和SP;电针组在MCAO手术同时进行电针治疗;SP+电针组在电针治疗基础上于MCAO造模成功后侧脑室注射SP。以Zea Longa 6级评分法进行行为学评分,氯化三苯基四氮唑(TTC)染色法测定脑缺血面积,免疫组织化学法检测脑组织SP表达。结果与模型组和生理盐水组比较,电针组在MCAO后24 h神经行为学评分显著降低(P<0.05);SP+电针组神经行为学评分显著低于其他组(P<0.01)。24 h后与模型组和生理盐水组比较,电针组和SP+电针组脑梗死面积显著缩小(P<0.01)。SP+电针组大鼠大脑皮质和海马中SP含量显著高于模型组(P<0.01和P<0.05)。结论侧脑室注射SP可增强电针治疗局灶性脑缺血的疗效。
Objective To discuss the role of lateral intracerebroventricular injection of substance P(SP) in the treatment of focal cerebral ischemia with electroacupuncture.MethodsAll 50 modeled Wister rats were randomly divided into the model group,normal saline solution group(NS group),SP group,electroacupuncture group(EA group) and SP and EA group(SP+EA group) and a control group was set additionally(each n=10).The rat model of focal cerebral ischemia was established by using middle cerebral artery occlusion(MCAO).The NS group and SP group were given lateral intracerebroventricular injection of NS or SP respectively after MCAO.The EA group was given electroacupuncture at the same time of MCAO.The SP+EA group was given intracerebroventricular injection of SP on the base of electroacupuncture after MCAO.The Zea Longa graded approach of 6 grades was used for neuroethology scoring.The cerebral ischemia area was detected by applying staining of triphenyl tetrazolium chloride(TTC) and immunohistochemistry was used to detect SP expression in brain tissue.ResultsCompared with the model group and NS group,the neuroethology scores decreased significantly in the EA group 24 hours after MCAO(P0.05),and were significantly lower in the SP+EA group than those in other groups(P0.01).Compared with the model group and NS group,the cerebral ischemia area was reduced significantly in the EA group and SP+EA group 24 hours after MCAO(P0.01).The content of SP in cerebral cortex and hippocampus were significantly higher in the SP+EA group than those in the model group(P0.01,P0.05).ConclusionThe lateral intracerebroventricular injection of SP can improve the curative effect of electroacupuncture in the treatment of focal cerebral ischemia.
