摘要
采用RCS-585比色法对正常人尿中毒性羰基类物质(RCS)进行测定.最适反应时间和反应温度为40min,45℃,检测波长为585 nm.结果表明人尿中RCS在0.014~20μmol/L范围内呈良好线性关系:Y=0.11X+0.03,r=0.999 8,t=197.08,P>0.01;检测极限为0.014μmol/L,平均回收率为102.57%.对低、中、高3种浓度的尿液进行测定,变异系数均小于5%,表明精密度良好.此方法操作简便、快速,结果准确;适用于人类尿液中毒性羰基类物质的含量测定;对于检测人体的羰基应激和亚健康状态有很好的临床或临床前实用价值.
The toxic reactive carbonyl substances(RCS) in urine of the healthy people were measured by an improved RCS-585 method.The optimum reaction period and temperature were 40 min and 45 ℃ respectively,with the detection wavelength at 585 nm.The results show that there is a linear range in 0.014~20 μmol/L RCS in human urine: Y=0.11X+0.03,r=0.999 8,t=197.08,P0.01.Using the method to detect toxic carbonyl substances in urine,the detection limit was 0.014 μmol/L and the average recovery rate was 102.57%.Three different urine samples were determined with RCS-585 method.The coefficient of variability were less than 5%.This method is simple,rapid and accurate.It could be used for detection of toxic carbonyls in human urine and may have important value for the detection of human carbonyl stress and sub-health state in clinic and pre-clinic situations.
