摘要
目的探讨白细胞介素(IL)-23、IL-17在实验性内毒素诱导葡萄膜炎(EIU)动物模型眼部表达的特点。方法 SD大鼠随机分为两组:正常对照组、EIU模型组。伤寒内毒素致敏诱导大鼠建立EIU模型,于建模后6、12、24、48、60 h用裂隙灯显微镜进行眼前节检查,并根据Hoekzema方法进行眼部炎症评分,RT-PCR法检测大鼠眼部IL-23 mRNA、IL-17 mRNA表达水平。结果 EIU组大鼠双眼在免疫后6~48 h均可见眼部炎症反应,最高眼部炎症评分为(5.40±0.44)。与对照组相比,EIU组大鼠葡萄膜组织中IL-23 mR-NA及IL-17 mRNA表达增加,差异有统计学意义(P〈0.05)。相关性分析结果显示,眼部炎症严重程度与IL-23mRNA及IL-17 mRNA表达呈正相关。结论 IL-23 mRNA及IL-17 mRNA参与EIU发病过程。
Objective To investigate the expression of IL-23 mRNA and IL-17 mRNA in endotoxin-induced uveitis(EIU) for discuss IL-23/IL-17 in EIU pathogenesis and the possible function mechanism.Methods SD rats were randomly divided into two groups,control group and model group.The models rats of severe EIU were induced by the plantar injection of salmonella typhimurium edotoxin.The clinical manifestation of eyes were observed under the slim-lamp and opthalmoloscope,and scored based on the Hoekzema′s criteria.The rats were sacrificed at 6,12,18,24,48 and 60 h after injection of endotoxin.The expression of IL-23 mRNA and IL-17 mRNA in ocular sections were detected by RT-PCR.Results After endotoxin injection,ocular inflammation scores increased during 6 h to 48 h.The highest ocular inflammation score was 5.40±0.44.IL-23 mRNA and IL-17 mRNA expression levels were significantly increased after injection.Absent IL-23/IL-17 mRNA expression was seen in the negative control sample.The intensity of IL-23/IL-17 mRNA expression was significantly positively correlated with EIU findings.Conclusion High level IL-23/IL-17 mRNA in EIU is associated with disease activity.
