外源性胰岛素对2型糖尿病大鼠抗炎及肾脏的保护作用 被引量:1

摘要 目的观察外源性胰岛素对2型糖尿病大鼠血、尿细胞间黏附分子1(ICAM-1)的表达及肾组织结构和功能的影响。方法 30只健康SPF级雄性SD大鼠,10只作为正常对照组(A组)喂饲常规饲料;20只采用喂饲高糖高脂饮食,3个月后一次性腹腔注射小剂量链脲佐菌素,建立2型糖尿病大鼠模型;模型大鼠随机分为糖尿病组(B组)和胰岛素治疗组(C组)。8周后检测血糖、血脂、尿视黄醇结合蛋白(URBP)、尿白蛋白肌酐比(ACR)及血ICAM-1(SICAM-1)、尿ICAM-1(UICAM-1)水平,留取肾组织用于病理学检查。结果 B、C组空腹血糖及糖化血红蛋白水平、SICAM-1、UICAM-1、URBP、尿ACR均高于A组,C组UICAM-1排泄较B组明显降低,且UICAM-1与尿ACR、URBP及肾脏肥大指数呈正相关;C组大鼠肾小球ICAM-1表达明显减少,肾脏病变程度较B组明显减轻。结论外源性胰岛素对2型糖尿病大鼠肾脏有一定的保护作用,其机制可能与其抑制肾组织ICAM-1表达及UICAM-1排泄有关。 Objective To observe the effect of exogenous insulin on the level of ICAM-1 in serum and urine and the change of renal structure and function of type 2 diabetic rats.Methods 30 healthy male SD rats of SPF grade were chosen.10 of them were randomly selected as normal group(group A) who fed with normal diet,while the others were established as type 2 diabetes rat model by using high sucrose-high fat diet and injecting low dose Streptozotocin.Then these rats were randomly divided into type 2 diabetes group(group B) and insulin-treated group(group C).After 8 weeks,blood glucose,serum lipids,URBP,UACR,serum ICAM-1(SICAM-1) and urinary ICAM-1(UICAM-1) excretion rate changes were examined,specimens of rat kidney were keeping for pathological examination.Results Compared with group A,the blood glucose,glycated hemoglobin,SICAM-1,UICAM-1,URBP,UACR in group B and group C were significantly increased,while the four indicators in group C were significantly lower than group B.In addition,the change of UICAM-1 was positively correlated with UACR,URBP and kidney hypertrophy index.The ICAM-1 expression was significantly reduced in group C compared with group B.Conclusion Exogenous insulin has reno-protective effect,which may be partly related to the inhibition of ICAM-1 expression and excretion of UICAM-1 in renal tissue in type 2 diabetic rats.
