摘要 一、microRNA概述 microRNA简称miRNA,是一种内源性非编码单链小分子RNA,大小约为22个核苷酸.在结构上,编码miRNA的基因由RNA聚合酶Ⅱ转录,只有少数由RNA聚合酶Ⅲ转录[1];接着原始转录本被RNA酶ⅢDrosha加工而产生Pre-miRNA:随后Pre-miRNA在Ran-GTPase依赖的转运蛋白exportin-5作用下易位到细胞质中[2],被另一个RNA酶ⅢDicer进一步加工,切割产生成熟的单链miRNA,其5'端最弱碱基配对区被优先装载入RNA诱导的沉默复合物(RISC)中,miRNA与靶标mRNA的结合发生在RISC内,并且靶标的沉默需要Argonaute蛋白质家族的存在;随后miRNA与靶标mRNA 3'端配对并沉默其翻译成蛋白质.基因表达的转录后调控是一种对中枢神经系统神经元发育和行使正常生理功能重要的调控机制,而miRNA的研究补充了这一调控,对神经系统疾病的研究进入了新的领域.
